Description:
Modified DNA Polymerase is an N-terminally truncated version of Taq DNA polymerase.
Enzyme catalyses polymerisation of nucleotides into duplex DNA in the 5'-3' direction in presence of Mg2+ ions as robust Taq Polymerase. Enzyme lacks 5'-3' exonuclease activity in comparison to its precursor.
This polymerase is more heat-stable than Taq polymerase and works in a more wide Mg2+ concentration range. This feature allows optimising reaction conditions easier than for other polymerases.
Storage and dilution buffer:
10 mM K-phosphate, pH 7.0, 100 mM KCl, 0.5 mM EDTA, 1 mM DTT, 50 % glycerol, 0.01 % Tween-20.
Unit definition:
One unit of activity is the amount of enzyme required to incorporate 10 nmoles of dNTP into acid-insoluble material in 30 minutes at 72 oC.
Amplification buffer 10x:
166 mM (NH4)2SO4, 670 mM Tris-HCl, pH 8.8 at 25 °C, 0.1 % Tween 20. Recommended Mg2+ concentration 2.5 mM.

PCR, long distance PCR (blend with proof-reading enzyme), High Fidelity PCR, primer extension.

Activity, SDS-PAGE purity, absence of endonucleases/nickases and exonucleases.
