Hybridoma clones have been derived from hybridization of Sp2/0 myeloma cells with spleen cells of Balb/c mice immunized with purified human PAPP-A/proMBP (heterotetrameric PAPP-A, htPAPP-A) complex or purified human atherosclerotic tissue form of PAPP-A (MAb PAPP52).

Specificity:
Human PAPP-A antigen purified from placental blood (heterotetrameric complex, consisting of PAPP-A subunits and proMBP subunits). MAbs 11E4 and 5H9 are specific to proMBP subunit.

MAb isotypes:
IgG1 for MAb PAPP52
IgG2a for MAbs 4G11, 10A5, 3C8, 10H9, 18A10 and 7A6
IgG2b for MAbs 5H9, 11E4, 10E1 and 10E2

Applications:
htPAPP-A and dPAPP-A (PAPP-A dimer) immunoassays and immunoprecipitation.

PAPP-A immunodetection in Western blotting. MAb 18A10 recognizes PAPP-A subunit in reducing and non-reducing conditions after antigen transfer onto nitrocellulose membrane after SDS-gel electrophoresis. MAbs 4G11, 10A5, 18A10 and 10E1 recognize htPAPP-A, consisting of PAPP-A subunits and proMBP subunits, in non-reducing conditions.

Purification:
Chromatography on protein A Sepharose.
