Monoclonal mouse anti-human C-peptide

MAbs:
1H8, 2B7, 4H8, 5B8, 7E10, 2A11

Hybridoma clones have been derived from hybridization of X63-Ag8-653 myeloma cells with spleen cells of Balb/c mice immunized with recombinant C-peptide construction.

Specificity:
No cross-reactivity with human, bovine, porcine and mouse/rat insulin.
Affinity (Kd) > 1 x 108 M for all MAbs.
All MAbs bind with equal efficiency to the human free C-peptide and C-peptide region in proinsulin molecules. Antigenic determinants identified by MAbs are in the three-dimensional structure generated by N- and C- terminal regions of C-peptide separated by beta-turn at position 47-50 of proinsulin.

MAb isotypes:
IgG1, kappa for all MAbs

Applications:
Competitive ELISA or RIA detection of C-peptide in human serum after immunospecific removal of proinsulin (with solid-phased 1D6 or 1D3 MAbs to human proinsulin).
Construction of a sensitive two-site sandwich EIA/ RIA immunosorbent assay for the measurement of total human proinsulin (including proinsulin convertion intermediates) in serum.
Specific immunosorbtional removal of total proinsulin and C-peptide molecules from serum samples (with solid-phase 1H8, 2B7 and 4H8) before the measurement of human insulin.
