Description:
E. coli UDG - recombinant form of the enzyme, which catalyzes the release of free uracil from uracil-containing DNA. UDG efficiently hydrolyzes uracil from single-stranded or double-stranded DNA, but not from oligomers (6 or fewer bases)
Storage and dilution buffer:
25 mM Tris-HCl, pH 8.0, 100 mM NaCl, 0.1 mM EDTA, 1 mM DTT, 0.5 % Triton-X100, 50 % glycerol.
Unit definition:
One unit of enzyme activity is the amount that catalyzes the release of 60 pmol of uracil-containing DNA measured by release of [3H]-uracil in a 50 µl reaction containing 0.2 µg DNA (8.7x104 cpm/mg) for 30 minutes at 37 °C.
Reaction buffer 10x:
0.5 M Tricine, pH 8.5, 10 mM MgCl2 or buffer for PCR or RT-PCR protocol.
Applications:
PCR, RT-PCR, site-directed mutagenesis, and as a probe for protein-DNA interaction studies.
Quality control:
Activity, SDS-PAGE purity, absence of endonucleases/nickases and exonucleases.
